 1). pH meters -  Objective:                      To demonstrate how to use a standard pH meter in the TRACES Centre and undergraduate laboratories. This process explains how to accurately test a solution's pH.  2. Scope: Digital pH meters in the TRACES Centre and the UG Laboratory are affected. These devices include (but are not limited to) Sartorius, Oakton, and Mettler-Toldeo.  3. Responsibility :   Designated persons in the research lab are responsible for training employees and students on this protocol and ensuring that it is followed. Standard operating procedure for pH meter

 1). pH meters

Objective: 

                    To demonstrate how to use a standard pH meter in the TRACES Centre and undergraduate laboratories. This process explains how to accurately test a solution's pH. 

2. Scope:

Digital pH meters in the TRACES Centre and the UG Laboratory are affected. These devices include (but are not limited to) Sartorius, Oakton, and Mettler-Toldeo. 

3. Responsibility

 Designated persons in the research lab are responsible for training employees and students on this protocol and ensuring that it is followed. It is the responsibility of selected persons (Staff or Students) to adhere to the procedure's guidelines.

 4. Equipment:

 • pH meter, digital

 • Double-junction Ag/AgCl electrode pH electrode 

• ATC probe (automatic temperature control) (if available) 

• Transfer utensils, pipettes, spatula or capillaries 

• Clean stir bars on a stir plate (mini and micro) 

• pH Standard Solutions, pH 4,7 and 10 buffers 


5. Standard Operating Procedure:

 • Press ON to switch on pH meter. The measuring annunciator displays in the LCD's top centre. To indicate automatic temperature correction, the ATC Indicator shows in the lower right-hand corner.

 • Before used, rinse the electrode with de-ionized or distilled water to eliminate any contaminants stuck to the probe body. Unless otherwise specified, keep the sample at 25°C. Unless otherwise specified in the monograph, dip the electrode as well as the temperature sensor into the sample. 

• When dipping the electrode into the sample, ensure that the sensor or glass bulb of the electrode is thoroughly immersed in the sample. Gently stir the probe in the sample to ensure that it is homogenous.

 • Give yourself some time to let the reading settle. 

• When the reading is steady, the READY annunciator illuminates whether the READY indication has been enabled.

 • Hold down the HOLD key to freeze the measured reading, then press ENTER to confirm the measurement. 

• Fill up the test data sheet or technical information sheet with the observed pH and temperature values. 

• Wash the electrode in de-ionized water after use and store the electrode in storage solution or pH buffer 5.00/5.01 or 7.00/7.01 buffer solution. 7. Precautions: 

• Care should be given when inserting or removing the capillary from its cavity to prevent fracturing it; take it out vertically. 

• Always check the silicone oil's hue. It is usually colourless. If the oil appears to be yellow or brown, it should be discarded. Also, put some new oil in that container.
